This study was carried out to evaluate the possible mechanisms through which the Moringa oleifera leaves aqueous extract reveals its hepatoprotective effects in alloxan induced diabetic rats. Eighty albino rats were assigned into four groups. The control group was given sterile saline. The second group was injected with alloxan (150 mg/kg BW) i.p. The third group was given Moringa oleifera leaves aqueous extract (250 mg/kg BW) orally daily. The fourth group was injected with alloxan as the second group and administrated Moringa oleifera leaves aqueous extract as in the third group. Alloxan induced degenerative changes in hepatic and pancreatic tissues, increased hepatic lipid peroxidation and gene expression of pyruvate carboxylase and caspase 3. However, it decreased the activities of hepatic superoxide dismutase and catalase and gene expression of glycogen synthase. In contrast, Moringa oleifera leaves extract prevent the alteration of histoarchitecture of hepatic and pancreatic tissues, normalized hepatic reduced glutathione concentration, the activities of superoxide dismutase and catalase and gene expression of glycogen synthase, while it reduced blood glucose level, hepatic lipid peroxidation and gene expression of pyruvate carboxylase and caspase 3. This study indicated that Moringa oleifera leaves aqueous extract can be used as a potent antioxidant and hepatoprotective agent.
