This study was conducted to investigate the chemopreventive effects of dietary Calebin-A, a curcuminoid, on differentiation of 3T3-L1 adipocytes and high-fat diet (HFD) induced obesity and hepatic steatosis. Potential mechanisms contributing to these effects were also elucidated.METHODS AND RESULTS: Calebin-A effectively and dose dependently suppressed accumulation of lipid droplets in adipocytes through the suppression of adipogenic specific factor peroxisome proliferator-activated receptor (PPAR)γ and fatty acid synthase and activated acetyl-CoA carboxylase. Dietary Calebin-A effectively decreased weight gain and relative perigonadal, retroperitoneal, and mesenteric fat weight in HFD-fed mice. Furthermore, Calebin-A markedly reduced hepatic steatosis and the serum levels of glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, total cholesterol, and triacylglycerol. This herbal extract combination is an effective weight loss supplement for overweight and obese subjects. The present study aimed to investigate the thermogenic potential of the LI85008F in high-fat diet (HFD)-induced obese Sprague Dawley rats.METHODS: Seven rats received a regular diet (RD), and twenty-one rats received a high-fat diet (HFD) for 56 days. On day 28, the HFD-fed rats were randomized into three groups (n = 7). Starting from day 29 through day 56, one HFD-fed group received daily oral gavage of 0.5% Carboxymethylcellulose Sodium (CMC) alone (HFD), and the remaining two groups received 100 and 250 mg/kg LI85008F (LI85008F-100 and LI85008F-250, respectively). Body weight, fat mass, fat cell size, liver weight, liver triglyceride were measured. The energy metabolism parameters were measured using indirect calorimetry. In serum, the metabolic and endocrine markers were analyzed. The adipogenic and thermoregulatory proteins expression in the white adipose tissue (WAT) were analyzed using an immunoblot assay.RESULTS: Supplementation with both doses of LI85008F significantly increased resting energy expenditure (REE) in the obese rats. The LI85008F-250 rats showed significant up-regulation of uncoupling protein-1 (UCP-1) expression, as compared with the HFD rats. LI85008F significantly reduced body weight gain, fat mass, fat cell size, liver weight, and hepatic triglycerides. Employing a combination of molecular docking, scoring functions, and molecular dynamics simulations, 267 compounds were screened by docking on Mpro crystallographic structure. Docking score and interaction profile analysis exhibited strong binding on the Mpro catalytic domain with compounds C(1E,6E)-1,2,6,7-tetrahydroxy-1,7-bis(4-hydroxy-3-methoxyphenyl)hepta-1,6-diene-3,5-dione) and C(4Z,6E)-1,5-dihydroxy-1,7-bis(4-hydroxyphenyl)hepta-4,6-dien-3-one as lead agents. Compound Cand Cshowed minimum binding score (-9.08 and -8.07 kcal/mole) against Mpro protein in comparison to shikonin and lopinavir (≈ -5.4 kcal/mole) a standard Mpro inhibitor. Furthermore, principal component analysis, free energy landscape and protein-ligand energy calculation studies revealed that these two compounds strongly bind to the catalyticcore of the Mpro protein with higher efficacy than lopinavir, a standard antiretroviral of the protease inhibitor class. Annexin-V staining was performed to test the translocation of phosphatidylserine to the outer side of the cell membraneas a clear indicator of apoptosis.RESULTS: Turmeric extract significantly reduced the viability of OVCAR-3 cells both in 24 and 48 hours of exposure. OVCAR-3 cells treated with IC50 concentration of turmeric extract for 24 hours. 82.60% of cells were viable. The percentages of dead, early apoptotic and late apoptotic cells were detected to be 0.80%, 9.70% and 6.90%, respectively. Untreated OVCAR-3 cells have migration ability.
