However, the systematic studies of polyphenols constituents and cytotoxic ability in fruit parts of pomegranates derived from different Chinese cultivars have not been studied yet. In this report, a validated and sensitive HPLC-DAD method and fluorescence spectrophotometric method was established for quantitative analysis of four polyphenols and total phenolic content (TPC) in fruit parts of pomegranates (including peels, flesh, seeds, juices and leaves) derived from five Chinese cultivars, respectively. HPLC analysis was performed on the YMC ODS-A C18 column with gradient elution of MeOH and 0.1 % TFA. Four polyphenols including gallic acid, ellagic acid, punicalagin A&amp;B and punicalin A&amp;B exhibited satisfactory linearity in the concentration ranges of 20-320, 39-624, 74-1184 and 38-608 μg/mL, respectively. The results demonstrated that the amounts of TPC and four polyphenols in different fruit parts of pomegranates varied significantly. Peels of Sour-YRP possessed the highest content of punicalagin A&amp;B (125.23 mg/g), whereas other three polyphenols exhibited only trace. Among the five Chinese cultivars, Sour-YRP contained the highest content of TPC (688.61 mg/g) and could be considered as the desirable botanical source to obtain polyphenols. It is also discovered that low-maturity pomegranate might possessed much higher TPC than high-maturity pomegranate. The optimized HPLC-DAD method could be used for quality control of different pomegranates by identification and quantification of its main polyphenolic components. Furthermore, the in vitro cytotoxicity of different pomegranates fruit parts to cancer cells was evaluated. Benzo(a)pyrene [B(a)P] and N-nitroso-tris-chloroethylurea (NTCU) were used to induce lung tumors, and PFE was given in drinking water to A/J mice. Lung tumor yield was examined on the 84th day and 140 days after B(a)P dosing and 240 days after NTCU treatment. Mice treated with PFE and exposed to B(a)P and NTCU had statistically significant lower lung tumor multiplicities than mice treated with carcinogens only. Tumor reduction was 53.9% and 61.6% in the B(a)P + PFE group at 84 and 140 days, respectively, compared with the B(a)P group. The NTCU + PFE group had 65.9% tumor reduction compared with the NTCU group at 240 days. Immunoblot analysis and immunohistochemistry were used to determine effect on cell survival pathways and markers of cellular proliferation and angiogenesis. Prior work reveals an increase in PSA doubling time (PSADT) in a single arm study of pomegranate juice (POM) in PCA patients (pts) with a rising PSA after local therapy. We sought to determine the effects of 1 gram or 3 grams daily POMx on PSADT in a similar but more inclusive population of men seeking to defer androgen deprivation therapy. METHODS: This multi-center, double bind phase II, dose-exploring trial randomized men with a rising PSA and without metastases to receive 1 gram or 3 grams of POMx, stratified by baseline PSADT and Gleason score, and with no restrictions for PSADT and no upper limit PSA value. Men were treated until progression or for 18 months. PSA levels were obtained every 3 months. This study was designed to detect a 6 month on study increase in PSADT from baseline on each arm. RESULTS: 104 patients were enrolled and treated for up to 6 (92%), 12 (70%) and 18 months (36%). Median PSADT lengthened in the Intent to treat population (96% white, median age 74.5 years, median gleason score 7) from baseline 11.9 (range 1.6-54.6) compared to 18.5 (2-1523) months after treatment (p&lt;.001).There was no significant treatment difference on PSADT between the dose groups (p=.920). Forty-three percent (41/95) of men had a post-treatment PSADT of more than 200% of baseline PSADT and no new metastases. Declining PSA levels were observed in 13 pts (13%) during the study. No significant changes occurred in testosterone in either group. Although no clinically significant toxicities were seen, mild to moderate diarrhea was seen in 8 percent of patients. CONCLUSIONS: POMx treatment significantly increased the PSADT by over 6 months in both treatment arms, with no effect on testosterone.
